Ceramide kinase regulates TNF-?-induced immune responses in human monocytic cells

چکیده

Abstract Ceramide kinase (CERK) phosphorylates ceramide to produce ceramide-1-phosphate (C1P), which is involved in the development of metabolic inflammation. TNF-? modulates inflammatory responses monocytes associated with various disorders; however, underlying mechanisms remain not fully understood. Here, we investigated role CERK TNF-?-induced monocytes. Our results show that disruption activity monocytes, either by chemical inhibitor NVP-231 or small interfering RNA (siRNA), defective expression markers including CD11c, CD11b and HLA-DR response TNF-?. data upregulates phosphorylation. Inhibition significantly reduced secretion IL-1? MCP-1. Similar were observed CERK-downregulated cells. phosphorylation JNK, p38 NF-?B was inhibition CERK. Additionally, NF-?B/AP-1 suppressed Clinically, obese individuals had higher levels PBMCs compared lean individuals, correlated their levels. Taken together, these suggest plays a key regulating human during stimulation. may be relevant target for developing novel therapies chronic diseases.

SARS-CoV regulates immune function-related gene expression in human monocytic cells.

Severe acute respiratory syndrome (SARS) is characterized by acute respiratory distress syndrome (ARDS) and pulmonary fibrosis, and monocytes/macrophages are the key players in the pathogenesis of SARS. In this study, we compared the transcriptional profiles of SARS coronavirus (SARS-CoV)-infected monocytic cells against that infected by coronavirus 229E (CoV-229E). Total RNA was extracted from...

Ca2+/Calmodulin-Dependent Kinase Kinase α Is Expressed by Monocytic Cells and Regulates the Activation Profile

Macrophages are capable of assuming numerous phenotypes in order to adapt to endogenous and exogenous challenges but many of the factors that regulate this process are still unknown. We report that Ca(2+)/calmodulin-dependent kinase kinase alpha (CaMKKalpha) is expressed in human monocytic cells and demonstrate that its inhibition blocks type-II monocytic cell activation and promotes classical ...

Differential regulation of CD44 expression by lipopolysaccharide (LPS) and TNF-alpha in human monocytic cells: distinct involvement of c-Jun N-terminal kinase in LPS-induced CD44 expression.

Alterations in the regulation of CD44 expression play a critical role in modulating cell adhesion, migration, and inflammation. LPS, a bacterial cell wall component, regulates CD44 expression and may modulate CD44-mediated biological effects in monocytic cells during inflammation and immune responses. In this study, we show that in normal human monocytes, LPS and LPS-induced cytokines IL-10 and...
